Many pipe cleaners. Think 10 per strand. Or large-gauge jewelry wire wrapped in string (embroidery floss). Or legos (and glue). Or polymer clay a la sculpey. Or, as I'm using this year, lots of hope ;) .

I used copper wire to prototype a pre-build model, and it is very stable and retains its shape well. Just don't sit on it :P

In the image gallery there are photos of my sister's model from past years, in which she coiled stiff paper for the backbone and used paper-covered foam core for the bases, with many wire supports. It has held up quite well and looks good.

Is a pre-build of restriction endonuclease foki necessary for all levels of competition (regional, state, national)?

I think so. I don't see anything in the rules that mentions it being different at different levels of competition.
