For future reference, this is technically not the place to ask for official rule clarifications, but according to the clarifications posted here and here, your statement about the prebuild should be correct. Any other structures (i.e. amino acids, highlights of specific sites on the protein, etc.) you decide to include on your model can be considered for extra bonus points, and for those, I recommend you read the papers that these proteins are based on (posted on the MSOE CBM website), because that can help you get started, and what you opt to do can be anything your mind so desires to do.

Since this event has three parts (Prebuild, Onsite Build, Test), how long would the test probably be? I'm focusing on the exam section of this event, but I was wondering if the test would take up the whole time block or it would be short enough that I could assist in the onsite build.
2019 Events: Anatomy & Physiology, Designer Genes, Forensics, Protein Modeling.

From what I remember, the test was only about 10 open-ended questions. So it's usually a pretty short test.

Yes, just cysteine does disulfide bonds. Methionine has a methyl group at the end that precludes that.

This is my fifth time coaching this event. The test is not really long, but sometimes there is a somewhat open-ended "essay" question that can take longer to write, especially at state and nationals. You can get old Nationals tests from past years for an idea as to the protein questions that will be asked. Your best source of info will be the cbm.msoe site, which did finally upload a lot of CRISPR video links and links to copies of the technical papers which describe the findings of the structure of the SpyCas9-AcrIA4 complex. They have helpfully highlighted important sections to read. I believe the red-crossed out sections are sections you can ignore for instructional purposes.
However, I know at our state event, they go off in their own directions as far as questions they ask, and often totally ignore the topic (such as CRISPR), and just ask protein-DNA-RNA questions.
Usually two people can work on the test, after the protein section to be onsite modeled is set-up and adjusted as needed, and the third person works on the onsite build.

Should I just be getting one protein chain (yes this was mentioned earlier, but that was about the CRISPR, not Anti-CRISPER protein)?
And of the chain, which part contains the nuclease?
And should I build the Viral RNA, Tracer and CRISPR RNA as side chains?

FYI "side chain" usually refers to the additions on amino acids, not pieces of protein, those would be something like "subunits" or "domains". I think it would be a good idea to build those additional parts as well

Anyone know if this is how they grade? What's that model he's holding, and where does that model come from? Also do any event supervisors here know exactly what the process of scoring is like?

I believe it is just the Anti-CRISPR for the prebuild.
